A new method for rapid detection of influenza virus: gold nanorod-based immunoassay by lateral chromatography

Influenza virus is a common respiratory infection that can cause fever, cough and runny nose. Influenza virus epidemics can pose a serious threat to public health.

Rapid detection of influenza viruses is essential to control the spread of the epidemic. Currently, commonly used influenza virus detection methods include nucleic acid detection and antigen detection. Nucleic acid testing has the highest sensitivity and specificity, but requires sophisticated laboratory equipment and techniques. Antigen detection is easy to perform but has relatively low sensitivity and specificity.

Fluorescence-based lateral chromatography immunoassays (LFI) are rapid viral detection platforms that are easy to use and cost effective.LFI platforms use fluorescent molecules to label antibodies, which produce a fluorescent signal when the antibody binds to viral proteins.

However, conventional LFI platforms suffer from insufficient sensitivity and specificity. Metal-enhanced fluorescence effect (MEF) is a technique that utilizes metal nanoparticles to enhance fluorescent signals.

Prof. Kim’s team developed a gold nanorod (GNR)-based LFI platform that utilizes the MEF effect to enhance fluorescence signals. The platform uses Cy5 as a fluorescent molecule and binds it to GNRs, which have a plasmonic resonance effect on their surface that enhances the fluorescence intensity of Cy5.

The researchers conducted a series of experiments on the Cy5-mSiO2@GNR probe to study its fluorescence behavior. They found that the probe’s fluorescence intensity was highest when the thickness of the mSiO2 shell was 10.3 nm.

The optimized probe was integrated into the LFI platform to detect influenza A virus (IAV) at 1.85 pfu/mL. The sensitivity of the platform is much higher than that of the conventional LFI platform. In addition, the platform is highly specific for IAV and can effectively detect IAV even in the presence of other viruses.

The results indicate that the gold nanorod-based LFI platform is a promising method for the rapid detection of influenza viruses. With the advantages of high sensitivity, high specificity and simplicity, the platform can be used for early detection and screening of IAV and other viruses.
